凝血酶是一种多功能丝氨酸蛋白酶。它在炎症过程中产生,并诱导多种细胞的增殖和趋化,包括间皮细胞(mtc)。mtc是起源于中胚层的上皮细胞,血管内皮细胞也是如此。由于凝血酶作用于内皮细胞产生血小板活化因子(PAF)和内皮素(ET)-1,故推测MTCs也通过凝血酶的作用产生PAF和ET。将大鼠胸膜MTC (RMTC, 4/4 R.M.-4)单层膜在组织培养皿中培养不同时期。用高效液相色谱法分离上清,测定ET亚型和PAF物种。用酶联免疫吸附法测定免疫反应性ET,用血小板聚集仪测定PAF。在rmtc条件培养基中检测到ET-1、ET-2和ET-3,主要亚型为ET-1和ET-2。rmtc主要向上清中释放C16:0 PAF。通过凝血酶的作用释放免疫反应性ET和PAF。 Synthetic PAF significantly induced secretion of ET, but the PAF receptor antagonists, WEB2086 and E6123, failed to modulate thrombin-induced ET release. These results indicate that thrombin acts on pleural rat mesothelial cells to release ET and PAF, which may play a role in the development of pleurisy.
